Abstract: 

Electrorotation (ROT) is a powerful label-free electrokinetic technique for probing single-cell dielectric properties, yet its adoption remains limited due to the lack of standardized platforms, complex signal generation, and non-automated data analysis workflows. These gaps hinder systematic dielectric phenotyping and limit the translation of ROT into biologically relevant workflows. Within this context, we present the automatic Electro-Cell Physiometry (ECP) Platform, a ROT system that integrates hardware, software, and experimental protocols into a unified and user-independent system. The platform enables robust and reproducible extraction of complete dielectric spectra through fully automated image-based analysis. Experimental validation on heterogeneous cell populations shows high sensitivity in resolving subtle dielectric differences. These results support the transition of ROT from a specialized technique to a scalable and standardized tool for single-cell analysis.
